Genetic Determination of Tyrosinase Thermostability

چکیده
(HOROWITZ and SHEN 1952). The strain produces almost no tyrosinase activity when cultured at 35" C on a medium which favors the production of strong activity in 25" cultures. The evidence indicated that the temperature effect is not due to formation of a tyrosinase inhibitor, but to a net decrease in tyrosinase synthesis at the higher temperature. Attention has been called to the resemblance between this case and that of the Himalayan rabbit, in which the formation of melanin, the end-product of tyrosinase activity, fails to take place at body temperature (HOROWITZ 1951). On the basis of the analogy with Himalayan, a search was made for strains in which tyrosinase is produced at both 25" and 35" (analogous with selfcolored) and strains in which the enzyme is produced at neither temperature (analogous with albino). A number of strains analogous with self-colored, in the abovq sense, were found. When the tyrosinase in crude extracts of one of these strains was tested, it was found to be much more stable to heat than is the tyrosinase obtained from strain 4A. This discovery led us to an investigation of the following points : (a) the mode of inheritance of the thermostability, (b) the chemical basis of the difference in thermostability, and (c) the relationship between thermostability and enzyme production at 35". I' 4A of Neurospora c-yL1ssa is influenced by the temperature of cultivation

Tyrosinase Inhibitory Activity and Thermostability of the Flavonoid Complex from Sophora japonica L (Fabaceae)
Purpose: To investigate the tyrosinase inhibitory activity and thermostability of weak acid-treated Sophora japonica L. flavonoid complex (SJ-FC) in different solutions. Methods: The flavonoid complex of S. japonica was isolated and treated with weak acid to generate SJ-FC. The anti-tyrosinase activities of SJ-FC and well-known tyrosinase inhibitors were compared by mushroom tyrosinase activity...
متن کاملComparison of amino acid sequence and thermostability of tyrosinase from three wild type strains of Neurospora crassa.
The thermostability of tyrosinase from three wild type strains of Neurospora crassa has been investigated. For this purpose a sequence comparison of two thermostable and one thermolabile tyrosinase isoenzyme was carried out. It revealed that at position 201 the thermostable enzyme forms share an aspartate residue in contrast to an asparagine residue in the thermolabile form. In addition, one of...
متن کاملDetermination of the Thermodegradation of deoxyArbutin in Aqueous Solution by High Performance Liquid Chromatography
Tyrosinase is the key and rate-limiting enzyme responsible for the conversion of tyrosine into melanin. Competitive inhibition of tyrosinase enzymatic activity results in decreased or absent melanin synthesis by melanocytes in human skin. DeoxyArbutin (4-[(tetrahydro-2H-pyran-2-yl)oxy]phenol), a novel skin whitening agent, was synthesized through the removal of hydroxyl groups from the glucose ...
متن کاملDetermination of anti-melanogenic activity of Phlomis kurdica in human melanoma SKMEL-3 cells
The present study was designed to investigate the anti-melanogenic and cytotoxic activities of methanol extract of Phlomis kurdica. The antioxidant and anti-tyrosinase activity of MeOH extract from P. kurdica (MPk) were examined by DPPH radical scavenging and mushroom tyrosinase activity assays (in vitro), respectively. Furthermore, the effect of MPk on the melanin content, cellular tyrosinase ...
متن کاملIdentification, Cloning and Structural Analysis of Major Genes from Portulaca oleracea L. Hairy Roots that Involved in the Biosynthesis of Dopamine
Dopamine is one of the important medications of Portulaca oleracea L. To optimize the production of dopamine, one of the methods is the identification and engineering of metabolite pathways. To investigate the tyrosine decarboxylase (TDC) and tyrosinase, which seem to be the most important genes in dopamine synthesis pathway, hairy roots were produced from Portulaca oleracea using Agrobacterium...
